Two-dimensional cell culture (2D Cell Culture) is one of the most fundamental and widely used in vitro models in immune cell research. In this model, tumor cells or other target cells are typically cultured as a monolayer in standard culture dishes or plates. According to the research purpose, immune cells such as T cells, NK cells, macrophages, and dendritic cells can be introduced to investigate interactions between immune cells and target cells under controlled in vitro conditions.

 

Traditional 2D models have several advantages, including well-established culture conditions, simple operation, relatively short experimental periods, low cost, and compatibility with techniques such as fluorescence microscopy imaging, flow cytometry analysis, and high-throughput screening. Therefore, they have been widely used for studying immune cell functions and tumor immunology research for many years.
