Extracellular vesicles (EVs) are widely present in various biological fluids including blood, urine, saliva and cerebrospinal fluid. They carry diverse bioactive molecules secreted by parent cells, such as proteins, lipids, RNAs and DNAs. These molecular signatures are closely correlated with the physiological status and disease-associated alterations of the source cells, making EVs an important liquid sample for acquiring disease-related molecular information.Compared with conventional tissue biopsy, fluid-based EV detection offers advantages including relatively easy sample collection, minimal invasiveness and repeatable sampling. It enables capture of molecular signals associated with disease initiation, progression and therapeutic response from peripheral body fluids. Accordingly, EV-based liquid biopsy has become a key research area for precision diagnosis of cancer and other diseases.
Extracellular vesicles (EVs, including exosomes) derived from biological fluids. EVs can be isolated from body fluids such as blood, urine, saliva and cerebrospinal fluid. The vesicles are loaded with molecules including proteins, lipids, various RNAs (miRNA, mRNA, lncRNA) and DNA. EVs can reflect cellular physiological and disease status. Their lipid bilayer protects the cargo and maintains stability in body fluids, and the abundant molecular information facilitates detection and analysis.
EVs in different body fluids differ in cellular origin composition and molecular characteristics, so appropriate sample types can be selected according to research objectives. Blood is suitable for EV analysis of systemic diseases and tumors; urine applies to urinary system and metabolism-related research; saliva features convenient and non-invasive sampling; cerebrospinal fluid can be used to obtain EV molecular information associated with central nervous system diseases.
