Skeletal stem cells (SSCs) and skeletal stem and progenitor cells (SSPCs) represent key cell populations for bone formation, maintenance and regeneration. As an essential source of osteolineage cells, SSCs/SSPCs possess self-renewal capacity and multipotent differentiation potential. They can further give rise to osteoprogenitors, osteoblasts and other bone-related cell lineages, and play central roles in bone development, bone remodeling and injury repair.

In recent years, research on skeletal stem cells has gradually expanded from conventional cell characterization and in vitro differentiation studies to lineage tracing, stem cell niche regulation, bone defect repair and tissue regeneration mechanisms. Elucidating the fate determination, differentiation regulation and regenerative potential of skeletal stem cells enables in-depth understanding of bone reconstruction, and provides a critical theoretical foundation for the development of bone repair materials, cell therapy and regenerative medicine applications.
